BACKGROUND: Here we describe a new class of cryptides (peptides encrypted within a larger protein) with antimicrobial properties, named schistocins, derived from SmKI-1, a key protein in Shistosoma mansoni survival. This is a multi-functional protein with biotechnological potential usage as a therapeutic molecule in inflammatory diseases and to control schistosomiasis. METHODS: We used our algorithm enCrypted, to perform an in silico proteolysis of SmKI-1 and a screening for potential antimicrobial activity. The selected peptides were chemically synthesized, tested in vitro and evaluated by both structural (CD, NMR) and biophysical (ITC) studies to access their structure-function relationship. RESULTS: EnCrypted was capable of predicting AMPs in SmKI-1. Our biophysical analyses described a membrane-induced conformational change from random coil-to-α-helix and a peptide-membrane equilibrium for all schistocins. Our structural data allowed us to suggest a well-known mode of peptide-membrane interaction in which electrostatic attraction between the cationic peptides and anionic membranes results in the bilayer disordering. Moreover, the NMR H/D exchange data with the higher entropic contribution observed for the peptide-membrane interaction showed that schistocins have different orientations upon the membrane. CONCLUSIONS: This work demonstrate the robustness for using the physicochemical features of predicted peptides in the identification of new bioactive cryptides. Besides, it demonstrates the relevance of combining these analyses with biophysical methods to understand the peptide-membrane affinity and improve further algorithms. GENERAL SIGNIFICANCE: Bioprospecting cryptides can be conducted through data mining of protein databases demonstrating the success of our strategy. The peptides-based agents derived from SmKI-1 might have high impact for system-biology and biotechnology.

Objetivo: conhecer o cotidiano do adulto jovem com a doença renal crônica em tratamento hemodialítico. Métodos: pesquisa qualitativa, exploratória e descritiva, realizada com oito informantes em um serviço de nefrologia e no domicílio em um município no Sul do Brasil, no período de junho a agosto de 2013. Utilizou-se como referencial teórico, conceitos relacionados aos aspectos culturais de Madelaine Leininger e Clifford Geertz e os instrumentos para a coleta de dados foram a entrevista narrativa, a observação assistemática e o diário de campo. Resultados: os informantes estabeleceram uma nova história, devido à convivência cotidiana com a doença, apresentando modificações nos hábitos alimentares, hídricos, atividades diárias, laborais, lazer e sexual. A dependência de outras pessoas também foi mencionada. As fontes de apoio estiveram presentes nas narrativas. Considerações finais: os adultos jovens nessa condição de adoecimento construíram um novo modo de viver baseado em sua cultura, mantendo normas, valores e crenças.


Objective: to know the experience of a young adult with chronic kidney disease under hemodialysis.Methods: qualitative, exploratory and descriptive research, performed with eight informants in aservice of nephrology, and in a home in a city of south of Brazil, during June to August 2013. Thetheoretical referential concepts of Madelaine Leininger and Clifford Geertz was used, and theinstruments for the data collection were the narrative interview, the systematic observation, and afield diary. Data analysis was by Minayo. Results: the informants established a new history, due tothe everyday living with the disease, presenting changes in the habits of life. Dependence on otherpeople and sources of support such as family, people known or not, health team were mentioned.Final Considerations: young adults in this condition of illness built a new way of living, which isbased on their culture, keeping rules, values and beliefs.

The present study evaluated the impact that orchiectomy, a routine but painful intervention in bovine husbandry, can cause on pulmonary immunity. To identify whether orchiectomy can impair lung defense, analyses of serum cortisol concentration and of alveolar macrophage and their function (phagocytosis and respiratory burst) were evaluated. Sixteen Holstein bulls (7 mo old, 250±50kg of body weight BW) were divided in two homogeneous groups - the castrated group and the sham group - and the sample were collected on Days -7, 1 and 7 relative to the day of the procedure. Serum cortisol concentration statistically increased on Days 1 and 7 (D-7: 4,97±1,28ng/ml; D1: 6,35 ±1,10ng/ml; D7: 8,28±1,94ng/ml) after castration and these variables seem to impact the alveolar macrophage percentage on D1 (D-7: 76,86±3,44%; D1: 60,92±2,44%; D7: 74,17±2,56%) and their respective function of phagocytosis (P) and the oxidative burst (OB) on Days 1 and 7 for the castrated group (P D-7: 56,25±15,63 arbitrary values; D1: 54,75±14,07 arbitrary values; D7: 31,77±8,44 arbitrary values; and OB D-7: 222,34±39,52 arbitrary values; D1: 135,25±37,68 arbitrary values; D7: 117,73±18,17 arbitrary values). These results indicate that surgical castration affected lung defense until seven days after the practice, so the pulmonary cell function was impaired for a period higher than that reported in the literature.(AU)


O presente estudo avaliou o impacto que a orquiectomia, uma intervenção dolorosa comumente realizada durante a criação de bovinos, pode causar na imunidade pulmonar. Para tanto, foram realizadas dosagens de cortisol sérico, bem como a análise de macrófagos alveolares e suas funções (fagocitose e metabolismo oxidativo) de 16 bovinos da raça Holandesa preto e branco (sete meses de idade, 250±50kg de peso vivo). Esses animais foram divididos aleatoriamente em dois grupos homogêneos - grupo castrado e grupo controle - e foram avaliados nos dias -7, 1 e 7, relativos ao dia do procedimento cirúrgico, que foi realizado no dia 0. A concentração de cortisol sérico aumentou estatisticamente nos dias 1 e 7 em relação ao grupo controle (D-7: 4,97±1,28ng/mL; D1: 6,35 ±1,10ng/mL; D7: 8,28±1,94ng/mL). Notou-se diminuição de macrófagos alveolares no D1 (D-7: 76,86±3,44%; D1: 60,92±2,44%; D7: 74,17±2,56%) e de suas funções de fagocitose (F) e metabolismo oxidativo (MO) nos dias 1 e 7 (F D-7: 56,25±15,63 valores arbitrários; D1: 54,75±14,07 valores arbitrários; D7: 31,77±8,44 valores arbitrários; e MO D-7: 222,34±39,52 valores arbitrários; D1: 135,25±37,68 valores arbitrários, D7: 117,73±18,17 valores arbitrários) para o grupo castrado. Esses resultados demonstram que a orquiectomia afeta as defesas pulmonares por até sete dias após a prática, período superior ao relatado pela literatura.(AU)

RESUMO

Electrical injuries and especially those of high voltage still remain a source of high morbidity. Over the past few years, a change in the epidemiologic profile of these lesions was noticed at the Vall d'Hebron University Hospital Burn Unit, corresponding to an increase in cases out of the legal framework. It is our aim to describe this particular subset, to determine the extent of their injuries and to understand the reason for their increased incidence. We think this was favoured by the rise in the unemployment rate, along with higher copper prices.

Diversos fatores predisponentes são descritos para as afecções mamárias ou distúrbios secundários que comprometem a qualidade e produtividade de leite de fêmeas nas diferentes espécies. As características fenotípicas do úbere são consideradas na avaliação econômica de uma fêmea caprina, tanto pelo potencial de produção como pelo registro genealógico dessa fêmea. A limitação de estudos correlacionando essas características com a saúde do úbere gera a dúvida a respeito do significado da conformação do mesmo à saúde e produtividade da glândula mamária. Sendo assim, este estudo teve como objetivo relacionar os parâmetros da conformação do úbere com a celularidade da glândula mamária aferida pelo teste California Mastitis Test (CMT) e contagem de células somáticas (CCS) em 80 cabras da raça Saanen sem alterações no exame clínico da glândula mamária nem no teste de Tamis. Observou-se que a maioria dos parâmetros fenotípicos de úbere não influenciou a CCS, sendo considerados puramente estéticos. A circunferência e profundidade de úbere demonstraram correlação negativa com a celularidade e, por serem características de herdabilidade moderada a alta, podem ser parâmetros considerados para seleção genética de caprinos.


Several predisposing factors are described for mammary diseases or secondary disorders that compromise the quality and productivity of milk from females in different species. The phenotypic characteristics of the udder are considered in the economic evaluation of a female goat, whether for production potential, or as the genealogical record for these females. The limitation of studies correlating these features with the udder health raises doubt about the meaning of the conformation to the health and productivity of the mammary gland. Therefore, this study aimed to list the parameters of the udder conformation with the cellularity of the mammary gland checked by California Mastitis Test (CMT) and somatic cell count (SCC) in 80 Saanen goats without alterations in the clinical examination of the mammary gland or the Tamis test. It was observed that most of the phenotypic parameters of the udder did not influence the CCS, being considered purely esthetic. The circumference and udder depth showed negative correlation with the cellularity and moderate to high heritability traits can be considered parameters for genetic selection of goats.

Corynebacterium bovis is one of the most commonly isolated bacteria from aseptically collected bovine milk samples. The objective of the current study was to characterize the bovine innate immune response by evaluating milk polymorphonuclear neutrophilic leukocytes (PMNL) in mammary glands infected with C. bovis. Twenty quarters infected with C. bovis and 28 culture-negative quarters (with milk somatic cell count <1×10(5) cells/mL) were used. The percentages of milk PMNL and the PMNL expression of L-selectin (CD62L), ß2-integrin (CD11b), and one of the endothelial-selectin ligands (CD44), as well as the levels of intracellular reactive oxygen species (ROS) and the phagocytosis of Staphylococcus aureus, were evaluated by flow cytometry. The apoptosis and necrosis rates of the PMNL were quantified using dual-color flow cytometry with fluorescein-labeled annexin and propidium iodide. The present study revealed a higher percentage of PMNL in the milk from C. bovis-infected quarters, although no significant differences were found in levels of CD44, CD62L, or CD11b expression among the PMNL. A lower percentage of apoptotic PMNL was observed in C. bovis-infected quarters, as well as higher percentages of viable PMNL and of PMNL that produced intracellular ROS. However, no alterations were observed in phagocytosis of Staph. aureus by the PMNL or in intensity of intracellular ROS production by PMNL. Thus, results from this investigation of the PMNL function support, at least in part, the fact that intramammary infections by C. bovis may offer protection against intramammary infections by other bacteria.

Avaliou-se a confiabilidade e determinou-se a pontuação mínima relacionada à necessidade de intervenção analgésica de uma escala multidimensional para avaliação de dor pós-operatória em gatas submetidas à ovário-histerectomia. Trinta gatas foram filmadas em quatro momentos predeterminados: pré-operatório, pós-operatório antes e depois da analgesia de resgate e às 24 horas após a cirurgia. Quatro observadores, não cientes dos momentos de avaliação, e um observador, considerado padrão-ouro - pesquisador que desenvolveu a escala - , avaliaram os vídeos em ordem aleatória, a partir da aplicação da escala, e determinaram a suficiência ou não de analgesia, baseando-se na experiência clínica. Os vídeos foram reavaliados dois meses após a primeira análise. O grau de concordância entre as avaliações dos diferentes observadores e das avaliações feitas pelo mesmo observador em diferentes ocasiões foi verificado pelo coeficiente de correlação intraclasse, que variou entre bom e muito bom para todos os itens da escala, exceto atividade, atitude e miscelânea de comportamentos que apresentaram confiabilidade moderada. O ponto de corte ótimo (> 7) foi selecionado pela análise descritiva e da curva Receiver Operating Characteristic. Os resultados suportam a confiabilidade em termos de reprodutibilidade e estabilidade e indicam que a terapia analgésica deve ser considerada em pontuações >8.


The aim with this study was to evaluate the reliability, and to establish the minimum score relating to the analgesic intervention of a multidimensional scale for use in assessing postoperative pain in cats undergoing ovariohysterectomy. Thirty cats were video recorded at 4 predetermined moments: prior to surgery, before and after rescue analgesia postoperatively, and 24 hours after surgery. Four observers blind to the times of assessment, and a gold standard observer (researcher who developed the scale) evaluated the video segments in random order using the scale, and determined the adequacy or not of analgesia based on clinical experience. The videos were reassessed two months after the first analysis. The agreement between the assessment of different observers, and the assessment made by the same observer on different occasions has been verified by the intra-class correlation coefficient that ranged from good to very good for all scale items, except activity, attitude and miscellaneous behaviors, which showed moderate reliability. The optimal cutoff point (> 7) was selected based on descriptive analysis and Receiver Operating Characteristic curve. The findings support the reliability in terms of reproducibility and stability, and indicate that analgesic therapy should be considered in scores >8.

O manejo adequado da dor depende de métodos de avaliação acurados. O objetivo deste estudo foi avaliar a validade e a responsividade de uma escala multidimensional para avaliação de dor pós-operatória em gatas submetidas à ovário-histerectomia. Trinta gatas foram filmadas no período perioperatório em quatro momentos predeterminados: pré-operatório, pós-operatório antes e depois da analgesia de resgate e às 24 horas após o término da cirurgia. Cinco observadores não cientes dos momentos de avaliação e um observador considerado padrão-ouro (pesquisador que desenvolveu a escala) avaliaram os vídeos em ordem aleatória e determinaram os escores de dor dos animais, utilizando a escala a ser validada. A responsividade e a validade de conteúdo e constructo foram estabelecidas pelo aumento e pela redução significativa nos escores de dor, em resposta ao procedimento cirúrgico e à administração de analgesia de resgate, respectivamente, avaliados pelo teste de Wilcoxon (P<0,001). A validade de critério foi atestada pela concordância alta entre as avaliações do padrão-ouro com os demais observadores, analisada pela porcentagem de absoluta concordância e coeficiente Kappa ponderado. Os resultados suportam a validade e a responsividade de uma escala multidimensional para avaliação de dor aguda pós-operatória em gatas submetidas à ovário-histerectomia, quando utilizada por veterinários formados e com experiência clínica.


Adequate pain management depends on accurate assessment methods. The aim of this study was to evaluate the validity and responsiveness of a multidimensional composite scale for use in assessing postoperative pain in cats undergoing ovariohysterectomy. Thirty cats were video recorded in four predetermined moments during the perioperative period: prior to surgery, before and after rescue analgesia postoperatively, and 24 hours after the end of surgery. Five observers blind to the times of assessment, and a gold standard observer (researcher who developed the scale) viewed the video segments in a random order, and the cat's pain scores were registered using the scale to be validated. The content and construct validity, and the responsiveness were established based on significant increase and reduction in the pain scores, in response to surgery and analgesics administration, respectively (Wilcoxon signed rank test, P<0.001). Criterion validity was determined by high agreement between the evaluations of the gold standard with the other observers, assessed by the percentage of absolute agreement and weighted Kappa coefficient. The findings support the validity and the responsiveness of a multidimensional composite scale to assess postoperative acute pain in cats undergoing ovariohysterectomy, when used by veterinarians with clinical experience.

The influences of age in calves' immune system are described in their first phase of life. We hypothesized that variations that occur in the main mechanisms of lung innate response can help to identify periods of greater susceptibility to the respiratory diseases that affect calves in the first stage of their life. This study aimed to evaluate the innate immune system. Nine healthy calves were monitored for 3 mo and 8 immunologic evaluations were performed. Bronchoalveolar lavage samples were recovered by bronchoscopy. The alveolar macrophages in samples were identified by protein expression of cluster of differentiation 14 (CD14) and underwent functional evaluation of phagocytosis (Staphylococcus aureus stained with propidium iodide and Escherichia coli). Data was assessed by one-way ANOVA (unstacked and parametric) and the Mann-Whitney test (nonparametric). Functional alterations in CD14-positive phagocytes were observed, with punctual higher intensity of phagocytosis in the third week and its decrease starting at 45 d of life. A gradual increase in phagocytosis rate was observed starting at this date. It is concluded that from 45 d of life on, alveolar macrophages have less phagocytic capacity but more cells perform this function. We suggest that this occurs because lung macrophages of calves start to maintain their immune response without passive immunity influence. Until 90 d of life, calves did not achieve the stability to conclude the maturation of local innate immune response.

A apoptose de leucócitos polimorfonucleares (PMN) é um evento central no processo de resolução da inflamação. Sendo a contagem de células somáticas (CCS) um indicador da situação imunológica da glândula mamária, o presente estudo buscou esclarecer a influência que esses fatores têm um sobre o outro e sobre a evolução do processo inflamatório. Marcaram-se as amostras de leite com anexina-V, iodeto de propídeo (PI), anticorpo anti-CH138A. Encontrou-se correlação negativa entre apoptose de PMN e CCS, além de diferença estatística entre um grupo de alta CCS e um grupo de baixa CCS quanto à taxa de PMN viáveis, em apoptose, em necrose e em necrose e/ou apoptose. De modo geral, o grupo de alta celularidade apresentou menos CH138+ em apoptose e mais células em necrose ou viáveis do que o grupo de baixa celularidade. Conclui-se que apoptose de PMN e CCS estão relacionados, e que em mamas com CCS elevada este evento está diminuído. Apesar de haver maior disponibilidade de fagócitos para a defesa nessa situação, os efeitos anti-inflamatórios da apoptose também estão diminuídos, enquanto os efeitos pró-inflamatórios da necrose estão aumentados, o que pode colaborar com a cronificação da inflamação.


Polymorphonuclear leukocyte (PMNL) apoptosis is central to the successful resolution of inflammation. Since Somatic Cell Count (SCC) is an indicator of the mammary gland's immune status, this study sought to clarify the influence that these factors have on each other and on the evolution of the inflammatory process. Milk samples were stained with annexin-V, propidium iodide (PI), primary antibody anti-CH138A. Negative correlation between SCC and PMNL apoptosis was found, and a statistical difference between high SCC group and low SCC group was observed concerning the rate of viable PMNL, apoptotic PMNL, necrotic PMNL and necrotic and/or apoptotic PMNL. Overall, the high cellularity group presented lower proportions of CH138 + cells undergoing apoptosis and higher proportions of viable and necrotic CH138+ cells. Thus, it can be concluded that PMNL apoptosis and SCC are related factors, and that in high SCC, milk apoptosis is delayed. Although there is a greater amount of active phagocytes in this situation, apoptosis' anti-inflammatory effects are decreased, while necrosis' pro-inflammatory effects are increased, which can contribute to chronic inflammation.

Systemic lupus erythematosus (SLE) is a chronic inflammatory autoimmune disease and can affect several organs and systems. It is characterized by high production of autoantibodies against nuclear compounds. TLR7/8/9 are responsible for nucleic acid recognition and they trigger proinflammatory responses through activation of NK-kappaB and Type I IFN production, making a bridge between the innate and the adaptative immune systems. We analyzed the frequency of TLR7 rs179008, TLR8 rs3764880, TLR9 rs5743836 and rs352140 in 370 patients with SLE and 415 healthy controls from southern Brazil. All analyses were conducted with regard to gender and ethnicity. Genotypic and allelic frequencies were different for TLR7 rs179008 (0.253 vs. 0.163, p = 0.020 and p = 0.003, OR for T allele: 1.74 CI 95% 1.12-2.70) and TLR9 rs5743836 (0.174 vs. 0.112, p = 0.045 and p = 0.017, OR for C allele: 1.59, CI 95% 0.99-2.57) between European-derived female groups. A higher frequency was observed for the presence of Anti-SSa/Ro for TRL9 rs5743836 C allele carriers (0.228 vs 0.126, Bonferroni corrected p = 0.06). No statistical differences were found for TLR9 haplotypic analyses. We suggest that TLR7 rs179008 and TLR9 rs5743836 can be considered SLE susceptibility factors for women of European descent in our population.

The mannose-binding lectin gene (MBL-2) has emerged as a candidate for systemic lupus erythematosus susceptibility, but studies in Brazilian population have not been conducted. To examine potential associations of mannose-binding lectin alleles G57E, G54D, IVSnt5, R52C and R52H with susceptibility to and clinical expression of systemic lupus erythematosus in southern Brazilian patients, we conducted a case-control study with 327 consecutive patients with diagnosis of systemic lupus erythematosus and 345 healthy controls. Genotyping was performed by restriction fragment length polymorphism-polymerase chain reaction assay. A statistically significant difference in the frequencies of allele R52C was observed in European-derived systemic lupus erythematosus patients when compared with controls (9.6% vs. 3.3%, p < 0.001, odds ratio: 3.15, 95% confidence interval: 1.76-5.62, p < 0.05). The frequencies of alleles G54D and G57E were not different between European-derived systemic lupus erythematosus patients and controls. There were no differences between clinical and laboratory features in systemic lupus erythematosus patients according to the presence or absence of mannose-binding lectin allelic variants. These results support an increased risk of systemic lupus erythematosus in European-derived individuals carrying allele R52C. Patients carrying this allele have an approximately three-fold higher odds ratio of developing systemic lupus erythematosus when compared with controls. Our data do not support associations between the mannose-binding lectin allelic variants studied and clinical expression of systemic lupus erythematosus.

Alcohol has a variety of short- and long-term effects on cell-mediated and humoral immune response. Herein, we have characterized the impact of high-dose EtOH administration on phenotypic and functional features of murine APC subsets, including dendritic cell (DC), macrophages and B cells. Impaired cytokine synthesis and Leishmania-phagocytosis was observed in peritoneal macrophages following EtOH administration. Moreover, EtOH exposure led to decreased levels of splenic myeloid DC and increased percentage of macrophages with no changes in splenic lymphoid DC and B cells. Adverse effects of short-term EtOH administration also resulted in impaired OVA-endocytosis by DC and macrophages. In contrast, EtOH consumption upregulates OVA-internalization by B cells. These changes on APC hierarchy may play a role shifting the fate of the immune response after EtOH ingestion. In addition to an overall downregulation of Toll-like receptor-TLR-4 expression by splenic APC, a downregulation of TLR-2 expression in macrophages was observed. Moreover, EtOH exposure altered the expression of co-signalling molecules on splenic APC, downregulating CD40 on macrophages and upregulating CD80 on B cells, with no impact on DC subsets. The net result of changes in TLR-mediated and co-stimulatory signals may determine the altered immunological status induced by acute consumption of alcohol. A direct impact of high-dose EtOH administration in the activation status of splenic CD4(+) T cells was observed. Together, our results demonstrated that short-term high-dose EtOH administration has differential impact on APC populations, downregulating splenic macrophages and DC activity but up-regulating B lymphocyte function as APC, and ultimately yielding a micro-environment that led to increased activation of CD4(+) T cells.

Although it is known that Peyer's patches are the major inductive site for S-IgA production and B1 cells contribute to half of the IgA plasma cells detected in the gut lamina propria, the type of contribution of mesenteric lymph nodes to the process is still unclear. Cytokines such as TGF-beta, IL-10, IL-4, IL-5, and IL-6, are required to promote IgA class switching and IgA synthesis. Aging-related alterations in T and B cells and in cytokine production are already known. Some reports have also proposed that S-IgA production might be altered in aged animals. Herein, we investigated the role of MLN and aging in S-IgA production. Two- to 18-month-old BALB/c mice were used to evaluate aging-related alterations and MLN were removed to study its role in S-IgA production. We found that MLN are important, although not essential for S-IgA production. In addition, we showed that production of IgA-related cytokines are well preserved in MLN but not in PP of aged mice and that S-IgA levels are not affected by aging. Our results suggest that MLN may play a complementary role in S-IgA production mostly in aged animals.

The influence of sex hormones on carbohydrate and lipid homeostasis was studied by comparing the response to glucose or insulin infusion in male and female dogs. The levels of blood glucose, serum immunoreactive insulin (IRI) and plasma free fatty acids (FFA) were determined. The hyperglycemia induced by glucose infusion (10 mg min-1 kg-1) for 60 min in summer was significantly higher in male than in female dogs. This sex difference in the response was confirmed in experiments performed in winter, when blood glucose levels of both sexes in response to the infusion reached higher values than those found in summer. The increase of serum IRI in response to glucose infusion was similar in both sexes in the summer experiment, while in winter the increase was significantly reduced and delayed in males. Glucose infusion into male and female dogs induced no significant changes in plasma FFA concentration in experiments carried out in winter or summer. Insulin infusion (16.6 mU min-1 kg-1) caused hypoglycemia in both sexes, which was of earlier onset in the females. The antilipolytic effect of insulin occurred in both sexes and was also stronger in females. The data show sexual and seasonal differences in response by dogs to glucose infusion and emphasize the participation of sex hormones in the homeostasis of energy substrates.
